This is a good walkthrough of Civil 3D Sample Line mechanics that includes some of the the more advanced options and edit capabilities that can get lost in the shuffle.
Here's a tip. You may find it handy to keep really different Sample Lines in separate Sample Line Groups since you also often have to publish them separately anyway.

Best to always remember that when you create Offset Alignments you can also create slope-controlled Offset Profiles (or not) in recent releases of Civil 3D.
Your Alignment naming convention and Name Template Rules get to be pretty important if you employ lots of Offset Alignments and Offset Profiles.

My only addition to Ron's great walk-throught is that you usually want to create a named modelspace View when you get the desired rotated result.
That saves you the effort of having to work throught the whole process again and the named View provides more consistent results.
